Calcium (Ca2+) ion plays an important role in myocardial ischemic and reperfusion injury. In this study, we compared the effects of nifedipine, verapamil, and diltiazem, three Ca2+ channel blockers (CCB), on myocardial ischemia and reperfusion injury. The left coronary artery was occluded for 30 min followed by 3 h of reperfusion in anaesthetised dogs. Nifedipine (30 microg.kg-1), verapamil (0.1 mg.kg-1) or diltiazem (0.1 mg.kg-1) was infused during ischemia. Infarct size was assessed using the triphenyl tetrazolium chloride (TTC) staining method. Infarct size was decreased by each CCB in a dose-dependent manner. Moreover, CCB reduced infarct size significantly. The results of the pharmacological study suggest that nifedipine, verapamil and diltiazem may be beneficial in the treatment of acute myocardial ischemia and reperfusion injury.Tom Smith (radio personality)
